The extraordinary honour has been delivered following a poll of all 542 members of Team GB and comes after the triple Olympic champion carried the flag at the closing ceremony in Beijing four years ago.
Hoy will lead out the team when they become the last of the 204 competing nations to enter the arena at the opening ceremony.
Hoy was the unanimous choice of athletes in a number of the 31 teams polled by the British Olympic Association, and was declared the overwhelming winner.
The cyclist said he couldn't think of "a bigger honour for an Olympian to lead out the team to a home Olympics."
He added: "It's a once in a lifetime opportunity."
